Evaluating the genetic diversity of Erythropalum scandens based on using inter-simple sequence repeat markers

Erythropalum scandens Blume, an emerging medicinal plant with great potential for drug development, also possesses high edible value. In this study, we investigated the genetic diversity of germplasm E. obtained from different geographical locations using inter simple sequence repeat (ISSR) markers. For purpose, 18 ISSR primer pairs a distinct background and adequate polymorphism were selected. We established optimal ISSR–PCR reaction system (20 µL) following parameters: 1 µL DNA template (60 ng µL−1), 1.2 primers (10 µmol 10 MasterMix, 7.8 H2O. A total 183 loci amplified pairs, which 121 (66.12%) indicated polymorphism. Moreover, 34 germplasms exhibited similarity coefficients ranging 0.7104 to 0.9563, distances 0.0447 0.3420, Nei’s index 0.1946, Shannon’s information 0.2982, suggesting intraspecific diversity. UPGMA cluster PCoA analyses distinguished Guangxi those collected Guangdong, Hainan, Fujian, Guizhou. However, Mantel correlation analysis revealed that variation among was not significantly related distance. The wild cultivated can help guide variety selection breeding. Furthermore, present study affinities scandens. Overall, our findings lay foundation conservation utilization resources, identification classification varieties, improvement at molecular level.
